One method I use to speed up production on my webcomic illustrations is to make layers to isolate patches of color, then merging them for final export as a .png to place in the page framework. This image shows a drawing at the point where I have flattened a couple layers with each other to conserve space on my layers menu. Too many layers ends up being more time consuming to scroll up and down so once I am totally convinced I will need no more adjustment to a certain color, I will merge it with other color layers. Each color I place may have it's own color so I can change the transparency to 'LOCK TRANSPARENCY' on the layers panel. This allows me to be a bit less precise in my line stroke putting down a huge swatch of color, or applying a highlight without worrying about spilling over into another color. Layers are also great for applying undershadows or glow.
