When creating layers of color for panels in my webcomic, I save time filling in large swatches of color by first outlining a containing edge of a specific color. This will prevent the color from flood filling my entire canvas. Once I establish the boundaries where that color will be, I switch from the ink tool (hot key I) to the flood fill key (hot key F) and press my LMB inside the boundary I drew. This will quickly fill in the color so I don't have to draw all that color in while in ink pen mode. This technique usually leaves an outline after flood filling, but by switching back to ink pen (I), you only have one line to draw over to fill the area you needed.
