Is it possible to set a hot key for a particular custom brush? For example, there are several custom brushes and only one of them is used for color mixing.

not at the moment, unfortunately.
I set up several custom brushes in a toolbox (view - toolbox palette; mine is stickied to the left of the screen) when starting a new painting.
I have a couple painting brushes, a customized eraser, and a blur brush from the palette knife tool presets in my toolbox stack.
when you save your WIP file as a .ptg, the toolboxes are saved along.
that is the quickest way to access a specific custom brush or switch between customized tools on the go I have found so far.
